Abstract (EN)
Aim: The aim of this research project; The main source of the economy of the pure blood Arab horses (Equus ferus caballus) used in nonsteroidal antiinflammatory drugs Metamizol Sodium (MS) following intravenous and intravenous administration of the change of pharmacokinetics according to gender and routes of administration is to determine. Materials and Methods: In this study; 14 (7 female, 7 male) pure horse with an average live weight of 380-450 kg were used. Intravenous (i.v.) and intramuscular (i.m.) administration at a dose of 25 mg / kg, 0 minutes before administration and 15, 30, 45, 60, 90 minutes after administration at 2, 3, 4, 6, 8 and 12 hours. The analytes from the blood plasma were determined by HPLC and LC-MS/MS after an optimized SPE extraction. Significant pharmacokinetic parameters (EAA, Cmax, tmax, OKZ, t1/2λz) of each of the three metabolites were determined. Results: No side effects were observed in the animals treated during the study. The method used was validated, the curve of the calibration for all metabolites; It is linear between 0.01-100 μg/mL (r2≥0.999); recovery 82.7-98.5%; the lowest detectable limit was found to be between 1.43-4.08 and the determinable limit of 4,11-13,61 μg/mL. The highest Cmax value for MAA is i.v. 114,40 and EAA0-12 in 374,11 μg/mL, the lowest in females i.m. Cmax 56,65 and EAA0-12 156,45 μg/mL were measured. AA for men in the highest i.v. 17.57; EAA0-12 i.m. 58.81 μg/mL, the lowest Cmax females i.m. 14.85 and EAA0-12 56.71 μg/mL. AAA for females i.v. 0.68, the lowest value in men i.m. 0.33. The tmax value for MAA is the earliest as i.v. 0.25 h, the longest men i.m. 0.70 h. AA is the smallest for men i.v. 0,75 h, i.m. 1.50 h after the application. The smallest value for AAA is i.v. After the application, it was measured as 1,00 h in men. The highest EAA0-12 females for AAA i.v. The lowest value of 2,47 μg/mL in men i.m. 1.73. Conclusion: In this study, we compared the pharmacokinetic parameters following i.v. and i.m. administration of MS in the recommended doses of NSAIDs commonly used in horses and MAA, which has an active metabolite, has reached an earlier and greater concentration in plasma than others. This method has allowed us to analyze the metamizole metabolites at very low limits in a short time. As a result of the analysis, it was determined that the blood level of all metabolites and the calculated parameters varied according to both the sex and the routes of application of the horses.
