Determination of soil borne bacterial diseases on tomato plants grown in greenhouses of aydin province

Abstract (EN)
Through this study, tomato plants grown in greenhouses of Aydın province were investigated to determine Clavibacter michiganensis subsp. michiganensis, Pseudomonas corrugata and soft rot Erwinia sp. in 1999-2000. No bacterial pathogen was found in tomato plants from 89 greenhouses examined in summer 2000. Eight out of 91 tomato growing greenhouses were found to be infected with bacterial disease in surveys carried autumn 2000. According to laboratory examinations, three Cmm were isolated from one greenhouse in Çine and two greenhouses in İncirliova. All Cmm suspected isolates showed the same bio- chemical characteristics as the know Cmm isolate (Cmm-9) and were found pathogenic on tomato plants (cv H2274). According to survey in tomato plants grown in greenhouses of Aydın, Pseudomonas corrugata and soft rot Erwinia sp. were not found in Aydın province. Four semi selective agar media (D2an, SCM, mSCM and KBT) were evaluated for the recovery and visible colony forming time of Clavibacter michiganensis subsp. michiganensis (Cmm) and the selectivity of unidentified bacterial isolates from tomato seed and plants. Recovery of Cmm on D2an, SCM, mSCM and KBT in comparison to nutrient dextrose agar (NDA) medium ranged from 97.3%, 20.0%, 107.0% and 64.0%, respectively. Percent reduction of unidentified bacteria was 36.4% on D2an, 27.3% on SCM, 63.6% on mSCM and 9.1% on KBT. Visible colonies of Cmm were observed after 3 days on KBT and NDA, 4 days on D2an, 6 days on mSCM and 9 days on SCM medium. The dyes (orange g, alizarin red, berberin sulfate, nile blue, eosine g, thionin, methyle orange, kresol rot and crystal violet) were also tested for inhibitory effect and colony differentiation of Cmm and the unidentified bacteria. Nile blue and crystal violet inhibited Cmm at 100*g/ml concentration but none of the dyes induced pigmentation of Cmm colonies
