Isolation of lactic acid bacteria from different food samples of azerbaijan and investigation of bacteriocin production capacities

In this study, lactic acid bacteria were isolated from dairy products of different regions of Azerbaijan. However, the samples were counted by classical method and Tempo BioMeriux device. Isolated lactic acid bacteria were identified as biochemical and morphological. Spot-on lawn and 2 different agar well diffusion methods were used to determine the bacteriocin production ability of these bacteria. The best results were obtained by agar well diffusion method on MH agar. Antimicrobial activity was detected in 29 of 98 isolates. As culture dependent method, 16S rRNA gene regions of the isolates were amplified and plasmid DNA was isolated. Fluorescence in situ hybridization (FISH) experiments and total nucleic acid extraction were performed to determine the microbial diversity of the samples as culture-independent methods. Gene sequencing based on 16S rDNA from total nucleic acid extracts was performed using the Illumina MiSeq platform. Using SDS-PAGE method, protein profile of some isolates with and without antimicrobial activity was investigated.
