Abstract (EN)
Agriculture, especially in developing countries, has a very important strategic place in our country and in the world. A large part of the economy of many countries is covered by agriculture. In parallel with the popularity of agriculture, there is an increase in the use of pesticides. Those employed in agriculture prefer to use pesticides in order to get many and healthy products in a short time. However, pesticides and pesticide residues have many harms to the environment, along with living creatures. Beneficial microorganisms living in the soil interact with fungi and bacteria that are harmful to plants. As a result of this interaction, they contribute to plant development by establishing symbiotic relationships with plants without the need for pesticides. The current thesis study was based on the question of whether the isolate EU07 belonging to the Bacillus amyloliquefaciens (NCBI Accession: PRJNA743875 ID: 743875) can be used as a biological control by drenching into soil replacing of the of chemicals. Four different soil trials were set up at carried out studies in the frame of this thesis: control, positive control (treated with EU07), negative control (treated with F. oxysporum f.sp. radicis lycopersici) and treated (treated with both EU07 and F. oxysporum f.sp. radicis lycopersici). Tomato seedlings were simultaneously planted in each experiment. Metagenome analysis was performed on the soil samples collected on the 4th and 7th days of the experiments, and ICP-MS analysis was performed on the leaves samples at the end of the 7th day. The positive control showed the highest plant growth compared to control and inoculated pots. Yellowing, drying and blackening of the root crown rot were observed on the tomato leaves in the infected control. In the treated soil experiment, samples that were close to drying showed recovering and blooming with introducing of of EU07 isolate in to the soil. In accordance with the metagenome analysis, beneficial microorganisms (such as genus belonging to Acidobacteria etc.) ratio increased in the positive control and declined in the negative control. This increase also occurred in the treated soil, but not as much as in the positive control. According to the Icp-ms analysis, the element percentages in tomato leaves grown in the soils sprayed with EU07 isolate were higher than in tomatoes grown in other soils. According to the ICP-MS analysis, the element percentages in tomato leaves grown in the soils sprayed with EU07 isolate were higher than in tomatoes grown in other soils.
