Abstract (EN)
Arginine deiminase; it is a bacterial anti-tumor enzyme that hydrolyzes arginine to citrulline and ammonia. Arginine deiminase enzyme is used as an antitumor drug in the treatment of arginine auxotrophic tumors such as hepatocellular carcinoma and melanomas. Polyhydroxybutyrate (PHB); it is synthesized as a carbon and energy source under conditions where substances such as oxygen, nitrogen, phosphorus and nitrogen are limited. These structures are also stored granular polyesters. They are polymers that can be completely degraded by biological means. Trehalase is a glycoside hydrolase enzyme that catalyzes the conversion of trehalose to glucose. It has been detected in many prokaryotic living groups. These are bacterial metabolites. In this study, Gr (-) Escherichia coli, Pseudomonas aeruginosa, Ralstonia eutropha and Gr (+) Bacillus cereus and Staphylococcus aureus bacteria were used. The ability of bacteria to produce these metabolites; in the presence of 1% different carbon sources added to the phosphate buffer, it was studied comparatively for 24 hours, 37 ºC, 0 and 150 rpm shaking conditions. The results show higher arginine deiminase activity, PHB production and trehalase activity of Gram (+) bacteria than Gram (-) bacteria under static conditions. According to the results of the study, it can be said that pure different carbon sources can also be used in the production of these metabolites.
