Abstract (EN)
In this study, acridine derivatives have been synthesized and studied for their potential to inhibit the bacterial biofilm formation. The nucleotide responsible for the formation of biofilms in bacteria is the cyclic dimeric guanosine monophosphate (c-di-GMP), which interacts with acridine derivatives to form the G-quadruplex structure. In this way, the mechanism controlling biofilm formation can be blocked. Within the scope of the study, it was investigated whether the acridine derivatives interact with c-di-GMP, as well as the effect on the Bacillus subtilis strain, which has the ability to form biofilms, has been tested. The effect of the acridine derivatives on c-di-GMP was determined by means of monomeric GMP assays, and the decreaase in the resulting concentration of GMP were determined using ultraviolet (UV) spectroscopy and high performance liquid chromatography (HPLC). The effect of these derivatives on the formation of biofilm was determined by microtiter dish biofilm formation assay. The synthesized acridine derivatives are novel structures that are not present in the literature and interact with c-di-GMP resulting an inhibitory effect on biofilm formation of Bacillus subtilis strains.
