Abstract (EN)
Our study aimed to examine the gene polymorphisms (SNPs) of DRD2 (rs1800497), LEPR (rs1137100, rs1137101, and rs1805094), and OPRM1 (rs1799971) and determine the relationship between them and weight regain in obese patients who underwent bariatric surgery. Five hundred and forty-six patients (466 women, 100 men, BMI 35–40 kg/m2, BMI ≥40 kg/m2) who were followed up clinically at Istanbul Fatih Sultan Mehmet Training and Research Hospital enrolled in the study. One hundred and seventy-nine healthy individuals (100 women and 79 men, 18.5>BMI) have taken as a control group. Seven hundred and twenty-five blood samples total have been processed. After the nucleic acid extraction from blood samples, SNP genotyping was performed by using high-resolution melting curve analysis (HRMA). The obtained data were subjected to DNA sequence analysis. There was no significant difference between C and T allele frequencies of the DRD2 TaqI A1 polymorphism (p=0.1, OR=0.54, 95% CI=0.24-1.21). Allele distribution did not differ statistically for the LEPR K656N (p=0.51, OR=0.75, 95% CI=0.14-3.93), LEPR K109R (p=0.78, OR=0.77 95% CI=0.35-1.72) and LEPR K109R polymorphisms (p=0.32, OR=0.64 95% CI=0.37-1.12). The distribution of A and G alleles between the patient and control groups was statistically significant for the OPRM1 A118G gene variation (p=0.001, OR=2.99 (95% G.A.=1.45-6.15) It has determined that OPRM1 G allele carriers showed a 3-fold increased risk for obesity than the A allele carriers. Although it has been observed that the T allele of the DRD2 TaqI A1 (p=0.1) polymorphism and the G allele of the LEPR Q223R (p=0.08) variant decreases the risk for developing obesity, the results were not statistically significant.
