Determination of some biological activities of black rosehip (Rosa pimpinellifolia L.) fruit growning naturally in Bayburt region

Abstract (EN)

Rosehip is one of our plants that is widely used in many fields, especially in the food sector, from past to present. In this study, black rosehip (Rosa pimpinellifolia L.) belonging to the rosehip family, which grows naturally in the Bayburt region, was preferred. This species is also known as Rosa spinosissima. In this study, it was aimed to determine the total phenolic content, antioxidant properties, antibacterial activity, element profile and antiproliferative effects of MCF-7 cancer line at various concentrations of Rosa pimpinellifolia fruit samples. In the study, methanol extract prepared using ultrasonic assisted method was used. Total phenolic content of the fruit extract was determined by Folin-Ciocalteu method, antioxidant capacity was determined by ABTS and DDPH method, antibacterial activity was determined by disk diffusion method, elemental profile was determined by ICP-MS method and antiproliferative effect was determined by CVDK-8 test. The total phenolic content of the fruit extract was determined as 37.5±0.2 mg GA/g, the antioxidant activity value was determined as 32.06±1.1 mg TE/g by ABTS method and 23.47±1.6 mg TE/g by DPPH method. Five different Gram-positive bacteria (Bacillus cereus BC 6830, Enterococcus faecalis NCTC 12697, Staphylococcus aureus NCTC 10788, Staphylococcus aureus BC7231, Staphylococus aureus ATCC 25923) and five Gram negative bacteria (Escherichia coli NCTC 9001, Escherichia coli BC1402, Escherichia coli NCTC 9001, Escherichia coli BC1402, Salmonella typhimurium RSSK 95091 and Yersinia enterocolitica ATCC 27729) were used to determine the antibacterial effect, and it was determined that Rosa pimpinellifolia fruit extracts prepared at 500 µg/mL concentration did not have antibacterial effect against the selected target pathogen strains. In addition, methanol extracts of Rosa pimpinellifolia fruit were applied at different concentrations to the MCF-7 breast cancer cell line, and the effect was observed after 72 hours. Accordingly, the highest cytotoxic effect was detected at a concentration of 40 µg/ml.
