Abstract (EN)

In this research, we aimed to determine PAHs (p-hydroxybenzoic acide, naphthalene, phenanthrene and pyrene) degradation ability of 17 halophilic and halotolerant isolates and the methabolic pathway used by the isolates during the degradation.Morphological and cultural features, and growing conditions of these isolates were determined. The PAH degradation ways used by the isolates were also identified by using spechtrophotometric method. It was observed that the isolates use catechol 1,2 dioxygenase and/or protocatechuate 3,4 dioxygenase in intradiol ring cleavage. PCR amplification was carried to define the gene zones which codify dioxygenases of these isolates. The amplified products were also analysed on 1% (w/v) agarose gels, and possible gene zones of catechol 1,2 dioxygenase and protocatechuate 3,4 dioxygenase were determined. Nucleotide sequence analysis of these gene regions were performed.Concequently, it was found that C-24, C-27, C-37, C-41, C-43, C-46, C-50, C-51, C-52, CH isolates could degrade the PAHs tested and these isolates could be used in bioremediation of the environments contaminated with PAH containing high salinity.
