Abstract (EN)
In this study, the effects of NaCl prepared at different concentrations ((0, 50, 100, 150, 200, 250, 300 mM)) on the growth of cucumber plants were aimed in in vitro conditions. The experiment was carried out in the plant tissue culture laboratory of Sivas Science and Technology University, Faculty of Agricultural Sciences and Technology. The experiment was conducted with 3 different cucumber varieties. MS was used as the basic nutrient medium in the study. Germination percentage (%), salt tolerance index (%), stem wet and dry weight (g), root wet and dry weight (g), actual water content (%), stem and root lengths and visual scale values, antioxidant capacity were determined by DPPH and ABTS method. When the effect of different media doses on the varieties was evaluated, the highest antioxidant capacity value was obtained from HD (173.21 μM TE g-1) after DPPH analysis and from HC (251.06 μM TE g-1 KM) after ABTS analysis. In the study, the highest germination percentage was obtained in variety 1 (94.36%) in terms of variety average and the highest germination percentage was obtained in HA (96.05%) in terms of medium average. In terms of actual water content, the variety average was highest in variety number 2 (91.16%) and the medium average was highest in HA (91.71%). The highest stem length was determined in cultivar number 1 (26.40 mm) and the highest average was determined in HA (24.48 mm) medium. Considering the results of the study, although there were differences among the varieties, the increase in NaCl concentration negatively affected germination and plant growth in cucumber.
