Abstract (EN)
In this research, production of Poly-ß-hydroxybutyrate (PHB) by Rhizobium strains which are isolated from the soil which is taken from different regions of Elazığ province and its districts in 1996 using ?Plant Infection Method? was studied. Rhizobium phaseoli CİAT 899 strains which is taken from Ankara Soil and Fertilizer Research Institute, is control group. Rhizobium strains were grown in Yeast Ekstract Mannitol (YEM) medium at 28 ºC temperature for 48 hours. After incubation period cultures were centrifugated and biomass which are obtained were dried. After dry cell weigh had been measured, PHB were obtained by ultrasonication. PHB is converted to crotonic asit by using sulfuric acid. The amount of the last substance is measurad by using UV spectrophotometer at 235 nm. According to Rhizobium phaseoli CİAT strain cell's dry weights, maximum PHB yields were obtained %23,10 percent in 200 rpm rinseing speed, %24,22 percent in %0,5 percent sugar's consantration and %29,58 percent after in 96 hours incubation time.According to be worked Rhizobium phaseoli CİAT strain cell's dry weights, PHB yields were obtained between %16,88 - %26,38 percent in different rinseing speeds , %16,08 - %26,84 percent in different sugar consantrations , and %2,53 - %31,16 percent in different incubation times. According to cell's dry weights in different rinseing speeds maximum PHB yield was obtained %26,38 percent in labelled 24F extract and in 200 rpm rinseing speed. According to cell's dry weights in different sugar consantrations maximum PHB yield was obtained, in labelled 23F extract, %26,84 percent in %0,5 percent sugar concentration. According to cell's dry weights in different incubation times, maximum PHB yield was obtained %31,16 percent, in labelled 24F extract, after 96 hour's incubation times.
