Micropropagation of some citrus genotyps with classical and new generation tissue culture techniques and determination of genetic stability

Abstract (EN)

In this study, 'Tuzcu 31-31' and 'C35 Citrange' rootstocks, which are important and widely used for citrus, were carried out in comparative propagation and rooting studies in the Plantform system and solid medium. First of all, micropropagation and rooting of citrus rootstocks were evaluated in solid MS and WPM media supplemented with BAP (0, 1.0, 2.0 mg / L), Kinetin (0, 0.5, 1.0 mg / L) and 2IP (0, 1.0, 2.0 mg / L) for micropropagation, in MS, ½ MS and WPM media supplemented with NAA (0, 0.5, 1.0, 2.0 mg / L) and IBA (0, 0.5, 1.0, 2.0 mg / L) for rooting. Based on the solid media, the best results in both genotypes were obtained from MS medium containing 2 mg / L BAP for micropropagation, ½ MS medium containing 0.5 mg / L NAA for rooting. Micropropagation and rooting studies were carried out in the Plantform system with the best-defined media content. As a result of the study, Plantform system showed better plant growth, multiplication coefficient and plant quality than solid culture system in Tuzcu 31-31 sour orange propagation medium. Plantform system in rooting medium was found to be more advantageous than solid culture nutrient medium in terms of root number, root length, plant height and rooting percentage. In the C-35 citrange , no statistically significant difference was observed between Plantform and solid culture in the replication medium, but better results were obtained in the Plantform system than the solid culture in terms of root length, plant height and rooting percentage. Genetic stability of plants grown and rooted in solid culture and Plantform systems was tested by SSR markers.
