The effect of nutrition restriction and starvation sirtuin and autophagia marker levels in placentals and fetus of pregnant mice

In general, fasting is the intake of little or no food in humans, ranging from 12 hours to three weeks. Food restriction; It is defined as the percentage reduction of the ad libitum diet (daily food intake) without changing all nutritional values. Autophagy is known to be stimulated to maintain cellular homeostasis in response to various stressful conditions such as food deprivation, oxidative stress and DNA damage. He described multiple roles for Sirt1 and Sirt2 in the regulation of autophagy.Sirt1 is known to directly affect autophagy by deacetylation of the essential components of the autophagy induction network, such as the products of autophagy genes (Atg) 5,7 and 8. In this study, mice were divided into 7 groups as control, fasting, S-24, S-48, 1W, 2W, 3W. The fetus and placenta were obtained from the mother on the 18.5 days of gestation and their weights and lengths were evaluated. Immunohistochemical expression levels of the genes such as SİRT1, SİRT2, ATG5 and LC3 / AB in organ-based and placental regions of the mice with food restriction and fasting were evaluated. Hematoxylin-eosin staining of fetuses and placentas was performed for morphological evaluation. According to the results of the experiment, it was found that nutrient restriction and various fasting applications were significantly lower between fetus and placenta weight and height measurements of maternal mice. There was no significant difference in the placenta according to immunohistochemical results in food restriction and fasting groups. However, significant findings were found in the organs of the heart and left extremity for SIRT-1 marker, intestine for ATG-5 marker and liver tissue for LC3 / AB marker.
