Abstract (EN)

Nanobiotechnology science; It is an interdisciplinary technology that gains importance due to its applications in medicine, biomedicine, space industry, engineering and imaging. Different chemical and physical methods are used for the synthesis of nanoparticles. Although the synthesis of nanoparticles made by these methods is produced in a short time and in high quantities with low costs, the nanoparticles produced have toxic properties because they undergo toxic processes during the production stages. The toxicities of these nanoparticles constitute a great disadvantage, as their usage areas increase and they easily enter the living spaces of people. Biologically synthesized nanoparticles provide advantages because they are biocompatible and environmentally friendly. In this thesis, Haloferax alexandrinus, which lives at high levels in terms of salt concentration, has a very low risk of contamination and is not a threat to human health, was used. For the production of silver nanoparticles, filtration and dialysis methods were tried for isolation with different salt and silver nitrate concentrations in the medium. ICP-OES, SEM-EDAX, TEM analyzes were performed to determine silver nanoparticles. The cytotoxic effects of the obtained AgNPs were investigated in SH-SY5Y neuroblastoma cell lines. As a result, AgNPs obtained from H. alexandrinus were found to be up to 20 µm in size, although most of them were 2-3 µm in size. The best medium for AgNP was obtained in 23% Salt solution and 0.05 mM AgNO3 concentration. The IC50 value of the obtained AgNPs was found to be >200 µg/ml after 48 hours of application on SH-SY5Y neuroblastoma cell lines.
