Abstract (EN)

This thesis study was conducted to develop a recovery protocol for appropriate vitrification and to provide solutions to the vitrification problem seen in many woody plant species by modeling it in Aronia melanocarpa through clonal micropropagation method in vitro conditions. In the study, vitrified explants were obtained by creating vitrification with different sucrose and different benzylaminopurine (BAP) concentrations using healthy clones, and then these explants were subjected to a vitrification termination protocol with a single dose of IBA, different sucrose and BAP concentrations, and the recovery process was monitored accordingly. The starting and ending times, rates and morphological observations of vitrification in Aronia explants, microscopic examination of stomata and morphological observations during acclimatization process were also included in the findings. As a result of the study, it was observed that vitrification was observed in 11% to 77% of the explants in VOPs containing BAP in most MS nutrient media. In the vitrified explants, a recovery rate of over 95% was observed on the 13th day in BAP-free MS nutrient media, while this rate remained between 26% and 66% in BAP-containing MS nutrient media. On the 31st day of VSP, the recovery rate in BAP-containing MS nutrient media varied between 66% and 94%. Thus, it was revealed in this thesis study that the vitrification seen in the explants in the clonal propagation of Aronia melanocarpa under in vitro conditions can be recovered by taking it into the vitrification termination protocol. This study also suggests ideas for solving the problem frequently encountered in other woody fruit species.
