Investigation of cytotoxic / apoptotic effects of biological synthesized nanoparticles on cancer cells

Abstract (EN)
Cancer is the disease with the highest mortality rate after heart diseases. Many treatment methods such as radiotherapy, chemotherapy, and immunotherapy are used in the treatment of this disease. With the developing nanotechnology, nanoparticles have been also used in cancer treatment. In this study, cytotoxic/apoptotic effects of gold nanoparticles and silver nanoparticles synthesized by B. adusta filtrate and extract were investigated. The cytotoxic/apoptotic effects of these nanoparticles on lung cancer (A549), colon cancer (HCT-116), liver cancer (Hep3B) chronic myeloid leukemia (K562) cells and healthy cell line lung epithelial cells (BEAS2B), were tested using trypan blue analysis and MTT assay. Concentrations in the range of 10-300 μg/mL were applied for Tripan mavisi analysis and the concentration at which the viability fell below 50% was determined as 10-100 μg/mL. Then, the concentrations of 10-100 μg/mL were applied and IC50 values were determined at 24, 48 and 72 h by MTT test. Based on the IC50 values determined by MTT test, caspase-3 activity of cancerous and healthy cells treated with AuNP and AgNP's were measured. The results of this study showed that AuNP and AgNP'shad a significant cytotoxic effect on the cancer cell lines.
