OBJECTIVE: To investigate the effects of melatonin, PGEi and E vitamine as antioxidants in experimental renal ischemia-reperfusion injury in rats histologically and biochemical. MATERIAL AND METHODS. 42 adult female Wistar rats with regular estrous cycles were randomly divided into six groups. Group 1 (n=7) was the control group; Group 2 (n=7): Only ischemia was produced in rats by clamping one renal artery for 60 minutes. Group 3 (n=7): Ischemia was produced in rats by clamping one renal artery for 60 minutes and reperfused for 60 minutes. Group 4 (n=7): Ischemia and reperfusion was produced for 60 minutes.Before fifteen minutes of reperfusion 20 mcg/kg/TM melatonin was performed. Group 5 (n=7): Ischemia and reperfusion was produced for 60 minutes.Before fifteen minutes of reperfusion 20 mcg/kg/TM PGEi was performed. Group 6 (n=7): Ischemia and reperfusion was produced for 60 minutes.Before fifteen minutes of reperfusion 500 mcg/kg/IM vitamin E was performed. For light and electron microscopic investigations renal samples put into %10 formaldehyde and gluteraldehyde solutions. Renal tissue and blood malondialdehyde (MDA) levels, superoxid dismutase (SOD), glutathione peroxidase (GSHPx) levels were determined. Kruskall Wallis analysis of variance was used in the statistical analysis of data. RESULTS: In experimental renal ischemia-reperfüsion injury in rats, the histological analyses of kidneys revealed that, the most extensive damage occured at ischemia-reperfusion group (p<0.02, Mann Whitney U test), which correlated with decrease in blood SOD, GSHPx and significiant increase renal tissue MDA concentrations. In spite of this, the histological and biochemical analyses of rats pretreated with melatonin showed the less damage (p<0.02, Mann Whitney U test). CONCLUSIONS." In our study, the histological and biochemical analyses revealed that; melatonin was more effective than PGEi and vitamin E as antioxidants, m experimental renal ischemia-reperfusion injury in rats to reduced the damage. Thus, melatonin may be the first preference in renal ischemia- reperfusion injury treatment.KEY WORDS.Renal ischemia-reperfusion, antioxidants, light microscopy, electron microscopy.
