The detection of prevalence of oncogenic human papillomavirus genotypes among women with cervical cancer and precancerous lesions in our region

Abstract (EN)

Cervical cancer is the second most common cancer among women worldwide. Human papillomavirus (HPV) is the primer etiologic agent of the cervical cancer. The aim of this study was to detect the prevalence of HPV genotypes among women with cervical cancer and precancerous lesions in Çukurova region. For this purpose, the samples from 27 women with squamous cell cervical cancer, 18 CIN1 (cervical intraepithelial neoplasia), 16 CIN2/3 and 122 women with normal cervical cytology as control group were investigated for HPV DNA by MY09/11 and GP5+/6+ consensus PCR (polymerase chain reaction) test. HPV genotyping was performed by the f-HPV typing Kit (Molgentix, Barcelona, Spain) which is multiplex flourescent PCR test. Fluorescent PCR products were analyzed by capillary electrophoresis on ABI 3130 genetic analyzer (Applied Biosystems) device. The prevalence of HPV was found to be 92.5% in cervical cancer, 81.2% in CIN2/3, %66,6 in CIN1 and 6.5% in the control group. In cervical cancer cases, HPV16 was the most common type with a rate of 66.6%, followed by HPV18 (11.1%) and HPV31, 58, 59 ve 68 (3.7%, each). The most prevalent type was also HPV16 (%56.2) among CIN 2/3 lesions and the next common types were HPV18 (12.5%), HPV11 (12.5%), HPV45, 51, 52, 58 ve 6 (6.2%, each). In CIN1 cases, the most frequently observed type was HPV59 (22%) and the other types were HPV16 (16.6%), HPV11 (11.1%), HPV18, HPV31 ve HPV 35 (5.5%, each). HPV16 and/or HPV18 infection was identified in 74% (20/27) of cervical cancer cases and in 62.5% (10/16) of CIN2/3 cases. In conclusion, the two most predominant types found in women with cervical cancer and CIN2/3 were HPV16 and HPV18 which are the types targeted by current prophylactic HPV vaccines. The investigation of the distribution of HPV genotypes in women with cervical cancer and precancerous lesions in our region is important for evaluating the impact of HPV vaccines.
