The cambial activity and molecular characterization of Corylus colurna L. growing in Bolu region

Abstract (EN)

This study was carried out to reveal the cambial activity and genetic diversity of Turkish Hazelnut (Corylus colurna L.) in Bolu ecology. In this study, in order to determine cambial activity, the same tree was visited in 8 different periods during the vegetation season and shoot samples were taken and cambial development was followed. 19 different Corylus colurna L. varieties and 5 different (Karafındık, Çakıldak, Yomra, Giresun Tombul Fındık, Karayağlı) standard cultivars (Corylus avellana L.) determined in Bolu province were tested for DNA by 5 iPBS retrotransposon markers. As a result of the study, it was determined that the new xylem was formed from the 1st period, and in the 3rd and 4th periods, it increased from 172 μm to 242 μm and the highest growth occurred in the 4th period. It was determined that cambial activity continued after these periods but the growth rate slowed down. A total of 85 loci were produced with five iPBS markers for the 24 isolates used in the study. iPBS markers clearly distinguish Corylus colurna L. and Corylus avellana L. groups. 76 (76.47%) of 85 amplification products obtained from iPBS - PCR were found to be polymorphic. The highest polymorphic band ratio was 88.89% for iPBS-2395 and the lowest rate was observed in iPBS-2378 primer with 44.44%. In the results of research; It was determined that cambial activity was the highest in Turkish Hazelnut in May and there was a slowing increase since June. In the study, iPBS retrotransposon markers were used for the first time in Turkish Hazelnut and it was found that it can be used successfully with its high polymorphism rate.
