Abstract (EN)
Brucellosis is an economically important zoonotic disease that is widespread all over the world. In our country the prevention has been made by vaccination of animals. For ewes Brucella melitensis REV-1 vaccine has been used. A more attractive vaccine with less side effect is need to be improved and should be applied to human. In this study it was aimed to determine a strong immune mediated Brucella vaccine by encapsulating the Brucellar outer membrane antigens with the optimum liposome formulation. In our study the outer membrane of the Brucella melitensis REV-1 vaccine strain was isolated and encapsulated with liposome and the humoral immune response in mice was investigated by examining the antibody titers with Wright agglutination method. The statistical analyses show that the Brucella outer membrane extract is successful for having humoral immune response in mice. Between the groups "Brucella outer membrane" and "Brucella outer membrane encapsulated with liposome" and between the groups "Live attenuated Brucella melitensis REV-1 vaccine" and "Brucella outer membrane encapsulated with liposome" statistically significant difference was determined in case of antibody titers. As a result it is thought that the results of our study that aimed to investigate humoral immune response in mice, have thought to be with the investigations that are trying to determine a subunit Brucella vaccine. However, the study has to be improved with different liposome formulations and different extraction methods of Brucella outer membrane.
