New biological markers in the diagnosis and follow-up of brucellosis

Abstract (EN)

Brucellosis is a common zoonotic disease all over the world, especially in developing countries. Brucellosis is mainly seen in two phases, acute and chronic. Numerous laboratory methods have been developed for the diagnosis of brucellosis, but there is still no special test that meets the criteria of high specificity and sensitivity, cheap and safe. In addition, it has not yet been discovered in a test to predict chronicity to evaluate response to treatment. Human ABI gene family member 3 (ABI3), Human C-type lectin domain family 12, member B (CLEC12B), Human DNA damage ınducible transcript 4 like (DDIT4L), Human zinc finger protein 1 (ZFP1) Human protein inhibitor of activated STAT4 (PIAS4), Human protein phosphatase 2a activator regulatory subunit 4 (PPP2R4), Human WD Repeat-Containing Protein 33 (WDR33) Human Indoleamine 2,3- Dioxygenase (IDO) are proteins that have recently been discovered and are involved in various stages of the immune system. This study was carried out in order to make important contributions to the illumination of brucellosis pathogenesis, to discover proteins that will show response to brucellosis treatment and that can predict chronicity. The study groups were formed into four groups as acute, chronic, subacute and recovering patients. Serums were separated from blood samples taken from patients in all groups, and protein levels in our study were determined with the Enzyme Linked Immunosorbent Assay (ELISA) method. It has been observed that the increasing levels of ABI-3, CLEC12B and PPP2R4 can be used to predict chronicity and the decreasing levels can be used to monitor treatment response, in contrast, the decreasing level of DDIT4L, IDO, PIAS4 and ZFP-1 can be used to predict chronicity and the increased level can be used to evaluate response to treatment. Besides, new clues were obtained in understanding the strategy of hiding Brucella bacteria from the immune system with the results of the study.
