Abstract (EN)
Budesonide, a glucocorticosteroid, is a broad spectrum of anti-inflammatory and antiallergic agent. It is used in the treatment of bronchial asthmatic COPD, Crohn's disease and ulcerative colitis. In this thesis, a new differential pulse voltammetry (DPV) method was developed and validated for the determination of budesonide in suspension ampoules, which is easy, fast, inexpensive, and environmentally friendly compared to previously available ones. The electrochemical behavior of budesonide was determined by cyclic voltammetry (CV) and DPV in different electrolyte and buffer solutions (pH 2 - 9). The analysis results showed that drug could be determined with the highest sensitivity in 0.1 M HCl solution containing 13% KCl and 8% methanol. The operating range of drug was determined to be 0.0 mV to -1400 mV. An irreversible well-defined reduction peak of about -859 mV was obtained. From the examination of pH-Ep graphs, it was observed that the peak potential shifted towards negative with the increase of pH. This showed that the reaction could occur with the addition of hydrogen. In the scanning rate study, the peak current increases with the increase scanning rate, and the potential shifted to more negative area. These showed that the reaction was adsorption controlled. In DPV, calibration curve was linear between 1.65 – 35.35 µg/mL. The limit of detection (LOD) and limit of quantification (LOQ) values were found as 0.52 µg/mL and 1.57 µg/mL, respectively. The developed method is sensitive and accurate, and can be successfully used for routine analysis in drug determination and quality control laboratories.
