Investigation of genotoxic, histopathologic and biochemical effects of butylparaben (butyl 4-hydroxybenzoate) on pubertal male rats

Abstract (EN)
İn this study, damages of using an organic compound Butylparaben (Butyl 4hydroxybenzoate) as an antifungal/antibacterial preservative in cosmetic products are being handled. Starting from the endocrine disrupting function, the genotoxic effects of butylparaben in pubertal male rats were measured by the Comet test as well as by histopathological, hematological and biochemical analyzes. For this purpose rats divided as positive control (n=3),negative control(n=6) and 6 dose groups(n=6).Then the dose groups divided again as 14 days and 28 days. During the experiment, animal body weights, food and water consumption were measured daily. When the experiment was ended, blood and liver specimens from animals were subjected to Comet test and measurement to detect DNA strand fractures. When the results were compared with negative control (corn oil) and positive control (methylmethanesulfonate) groups, DNA damage was observed to be increased in all dosing groups compared to the control group. With this info we concluded that butylparaben can cause a number of genetic distortions. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), glucose, creatinine, triglyceride and urea analyzes were performed in serum samples. Changes in ALT-AST and creatinine values were associated with liver and kidney damage. Hematological analyzes were performed on blood samples and values for dose groups were increased compared to the control group. Liver and kidney organ weights were measured and relative weights were calculated and compared, histological examinations were performed on tissue samples. The comparison of the observed changes with the negative control group showed that the butylparaben increased the congestion and degeneration in the organs.
