Evaluation of the genotoxic and cytotoxic effects of külsil fungicide in human peripheral lymphocytes used in Canakkale province

Abstract (EN)

In this study, in vitro genotoxic effects of Külsil, which includes triazole fungicide myclobutanil, was investigated by using chromosomal aberrations (CAs) and micronucleus test (MN) in human peripheral lymphocyte cultures. Effects of on mitotic index (MI) and nuclear division index (NDI) were also investigated for determination cytotoxicity. Lymphocytes were treated with 3,125; 6,25; 12,5; 25 and 50 ppm doses of Külsil fungucide for 24 and 48 hours. Külsil fungicide significantly increased chromosomal aberration frequency at all treatments except 3,125 ppm dose for 24 h when compared with the control and this increase was dose-dependent in both treatment times. In the same way, Külsil increased micronucleus frequency with dose-dependently at all treatments except 3,125 ppm dose for 48 h when compared with the control. Külsil fungicide was toxic on lymphocytes at 50 ppm with both treatment times. Külsil decreased nuclear division index but this decrease was not statistically significant. Külsil fungicide significantly decreased the mitotic index with dose-dependently at all treatments except 3,125 ppm for 24 h when compared with the control group. Most effective decreasing in mitotic index in 25 ppm treatment when compared with the control group in 24 h and 48 h were observed respectively 43,75% and 78,47%. The results of this experiments showed that Külsil fungicide, which includes triazole fungicide myclobutanil, has clastogenic and aneugenic effects on human lymphocytes cultures depending on dose.
