Effects of capsaicin on cell line of MDA-MB-231 and protective role of sodium selenite

Capsaicin is a strong, bitter, white, odorless, DMSO, toxic substance found as a active ingredient in hot pepper. Effects of capsaicin and sodium selenite active substances, which have previously been determined to reduce the growth rate of the tumor in cancer types such as stomach, colon, prostate; MTT, Comet, apoptosis necrosis, to the MDA-MB-231 breast cancer cell line, MDA and antioxidant enzyme activities have been investigated by applying the doses determined by the methods. The value of Capsaicin'in LD50 was found to be 50 μM. According to the results of cytotoxicity analysis, it was determined that cell viability decreased due to increased concentration of Capsaicin. According to the comet analysis for DNA damage detection in the cells, the control group, 25, 50, 100 μM Capsaicin and 5 nM sodium selenite in the groups applied DNA tail percentage, the, DNA damage was determined due to increased dose in tail length and tail moment values. After increasing doses of Capsaicin and sodium selenite, the highest number of apoptotic cells was reached at a concentration of 100 μM Capsaicin and 5 nM sodium selenite. The highest necrotic cell count was observed at a concentration of 100 μM Capsaicin and 5 μM sodium selenite in parallel with the number of apoptotic cells. Control and application groups for malondialdehyde (MDA) level and antioxidant defense system enzymes; superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities were evaluated in comparison with spectrophotometry. In the evaluations, the amount of MDA increased, while the decrease in antioxidant enzyme activities was observed. In light of these studies, Capsaicin'in human breast cancer cell line MDA-MB-231 caused toxic effects in vitro conditions in, sodium selenite, on the other hand, has been determined within the scope of the parameters that have been studied to reduce the reproductive rate by showing synergit effect with Capsaicin. The current study will contribute to the literature on this subject and will shed light on the comprehensive studies afterwards.
