Determination of the molecular characterization and biological activities of cyanobacteria isolated from lake Cernek

Abstract (EN)
Cyanobacteria are one of the important sources of known antioxidant and antimicrobial effects. In this study, cyanobacteria species obtained from Cernek Lake were isolated. Cyanobacterial strains obtained as a result of isolation were purified in BG-11 and BG-110 nutrient media. Phenotypic molecular identifications of the purified cyanobacteria species were made and their antimicrobial and antioxidant properties were investigated. Among the isolated cyanobacteria species, those with different profiles were determined, and species were determined by 16S ribosomal RNA analysis. In antimicrobial activity trials, Gram-positive (+) bacteria Bacillus cereus ATCC 7064, Micrococcus luteus NRLL 1018, Bacillus subtilis ATCC 6633, Staphylococcus aureus ATCC 25923, Gram-negative (-) bacteria Salmonella typhimurium LT2, Klebsiella pneumoniae ATTC, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, yeasts Candida albicans ATCC 14053 and Candida krusei ATTC 34135 were used. Microdilution (MIC-Minimum Inhibition Concentration) method was used in antimicrobial activity trials. In the MIC method, the concentration value of the last well in which growth did not occur is taken as a basis. Among all cyanobacteria extracts, the highest antimicrobial activity was seen in C8 and C30 coded cyanobacteria extracts. It was determined that the lowest antimicrobial activity among all cyanobacteria extracts was in the C15 coded cyanobacteria extract. In antioxidant activity studies, antioxidant activities were determined by looking at DPPH (2,2-diphenyl-1-picrylhydrazil) radical scavenging activities and Copper (II) ion reduction based antioxidant capacity (CUPRAC: Cupric Ion Reducing Antioxidant Capacity) measurement results of cyanobacteria extracts. Among all cyanobacteria extracts, the highest DPPH radicals scavenging activity was found in the C37 coded cyanobacteria extract, and the lowest DPPH radicals scavenging activity was found in the C48 coded cyanobacteria extract. When all cyanobacteria extracts were examined, it was determined that the highest CUPRAC value was in the C43 coded cyanobacteria extract, and the lowest CUPRAC value was in the C15 coded cyanobacteria extract.
