Investigation of apobec3b and apobec3g genes' expression levels and mutations in different breast cancer cell lines

Abstract (EN)

APOBEC enzymes are important players in immunity and may contribute to carcinogenesis. APOBEC3B and APOBEC3G belong to cytidine deaminases that able to mutate DNA and RNA by converting cytidine to uridine. We investigated the expression levels of APOBEC3B and APOBEC3G in four breast cancer cell lines (SK-RB-3, MCF7, MDA-MB-231, CRL 2329) using qRT-PCR and gene sequencing to detect mutation. APOBEC3B was highly expressed in MDA-MB-231, however, statistically not significant (p > 0,05). APOBEC3G was highly expressed in CRL 2329, however, no significance was found (p > 0,05). Also, two substitution mutations in APOBEC3B were detected which are (c.239A> G) in MDA-MB cell line and (c. 492A> C) in SK-BR cell line. No mutation was found in APOBEC3G. Understanding the molecular mechanisms of APOBEC3B and APOBEC3G; may give definitive knowledge about the molecular characteristics of the disease and have aided the crucial information about new source of mutation in breast cancers.
