Determination of cytocic activities of Cordyceps militaris (L.) link fungus on A549 lung cancer, C6 glioma cancer, and DU-145 prostate cancer cells

Abstract (EN)

Cancer is an important public health problem of our time. Target-spesific therapeutic potentials in research, mutation found in many different cells, may vary according to the general health status of the case. Understanding the different metabolic requirements of the various cells that produce an immune response against cancer has led scientists to the main goals. Modern phytochemical and pharmacological studies have shown that polysaccharides are one of the main bioactive compounds in fungi. Therefore, therapeutic alternative pathways should emerge that will advance the development of new anticancer agents. In this thesis study; The cytotoxic activity of DMSO extracts from Cordyceps militaris (L.) on A549 lung, DU-145 prostate and C6 glioma cancer cell lines was determined in vitro. In the study, mushroom sample no. A6848 taken from Istanbul, Şile, Kömürlük district was used. Cytotoxic IC50 values of mushroom extract was calculated statistically by cytotoxic activity MTT, 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H- (tetrazolium bromide) method. The cytotoxic activities of the extract varied depending on the dose. The highest cytotoxic activity in C. militaris extract on A549 lung cancer cell line was determined as 60.61% at a concentration of 2400 µg/ml. On DU-145 prostate cancer cell line, it was detected as 85.27% at a concentration of 2400 µg/ml. On C6 glioma cancer cell line, the highest cytotoxic activity was detected as 85.27% at a concentration of 2400 µg/ml. IC50 values on A549 lung, C6 glioma and DU-145 prostate cancer cell lines were calculated as 1343 µg/ml, 833.4 µg/ml and 723.2 µg/ml, respectively. It was determined that the mushroom extract showed the highest effect on the DU-145 prostate cancer cell line. The results show that DMSO extracts of the mushroom collected from the field have cytotoxic activity against cancer cell lines A549, DU-145 and C6. The data obtained in this study show that this mushroom extract have a high potential to be used in the development of drugs that can be used in the treatment and prevention of lung, prostate and glioma cancer. The results show that ethanolic extracts of this mushroom have cytotoxic activity against A549, DU145 and C6 cancer cell lines. The data obtained in the study reveal the potential of the mushroom extracts used in the development of drugs that can be used in the treatment and prevention of lung, prostate and glioma cancer.
