Investigation of ciprofloxacin nephotoxicity in rats

Whether oral ciprofloxacin will cause nephrotoxicity in rats, β-NAG, β2-Microglobulin, microalbumin, creatinine clearance, BUN, Na, K, pH parameters were investigated. Rats on days 0, 4, 7, and 14 were divided into 4 groups with 6 rats in each group. The 14th day group was formed from rats given ciprofloxacin on the 4th and 7th days and kept for 7 days. Rats were given 400mg / kg / day ciprofloxacin by nasogastric. The rats in each group were placed in metabolic cages at the end of the study day and 24-hour urine, blood and kidney samples were taken. In 24-hour urine samples, β - NAG (spectrophotometer), β2-Microglobulin (chemiluminescence), Microalbumin (radioimmunoassay), creatinine (autoanalyzer) were studied. Na, K, BUN, creatinine (autoanalyzer) were studied from the blood samples and creatinine clearance was calculated. β -NAG value of the group (Group 1) without ciprofloxacin is 7.8 ± 1.31 U / L, 27.1 ± 5.7 U / L of the group (Group 2) with ciprofloxacin for 4 days is 15.6 ± 2.1 U / L for the group given ciprofloxacin for 7 days. β-NAG value of the group (Group 4), where ciprofloxacin was given for 7 days and kept for 7 days, decreased to 7.5 ± 1.14 U / L. There was no significant change in other nephrotoxic parameters and kidney biopsies. These results show that ciprofloxacin causes nephrotoxicity in rats by causing early dose-free tubular damage. Another conclusion from our study is that β-NAG may be a marker that can be used in the early detection of ciprofloxacin nephrotoxicity. Our results emphasize that ciprofloxacin can cause nephrotoxicity and tubular damage plays a role in the mechanism of renal injury.
