Year:2©05,Pages:51 Jury: Assoc.Prof.Dr. Mukadder KAYIM Prof. Dr. Ali ERKTLIÇ AssocProf. Dr. Salih KAFKAS 4 Forty-six Fusarium şpp. isolates were obtained from randomly selected corncob, considered disease infected in Adana and Osmaniye provinces during the 2002-2003 years to determine fumonisin producing Fusarium species in the second crop maize in the year Çukurova plain. Microscopic observations showed that second crop maize was infected intensively with Fusarium moniliforme. It is concluded that this fungus produced high levels of fumonisins ranging from 0,1-7 ppm based on ELISA of maize kernels. While the highest average level of fumonisin was 3,4 ppm in Ceyhan- 1 region, the lowest average level of fumonisin with 0,35 ppm was determined in Kozan region. F. moniliforme species was determined by PCR analysis for a specific genomic DNA fragment amplification from other Fusarium species. The 1600 bp DNA fragment was amplified by PCR in all F. moniliforme isolates leading a selection from F. semitectum species. To identify genetic differences based on regional differences among the F. moniliforme isolates, RAPD analysis was applied and high level of genomic polymorphism was found among the isolates.
