Abstract (EN)

In addition to studies concerning genetic engineering which has gained importance in recent years, studies related to bioremediation of heavy metals causing serious environmental problems and extensive research on detoxification of xenobiotics has been started. Especially alternative new-use of bacteria species equipped with necessary genes that has the ability to break down substances to harmless compounds has lately been identified. For this purpose, with Vitreoscilla hemoglobin (vgb) cloned wild D. radiodrans gene recombinant and vgb¯was used as control recombinant strains. In this way, D. radiodurans' s Cd and Cr as toxic metal biosorption capacity and UV radiation resistance will be explored in addition, organisms will be exposed to more oxygenated environment by providing higher growth capacity with vgb gene,bacteria biosorption ability and UV resistance will be investigation is planned. At the same time,the effect of application of UV radiation to wild-type and recombinant D. radiodurans's antioxidant defense systems (superoxide dismutaz, catalase, and carotene) is explored and the impact on vgb effect on the defense system has been identified. In conclusion, aplication of high doses of UV-C treatment on vgb genes had a positive effect on bacterial reproduction potential and on enzyme activity after irradiation was determined. Also the effect of Cd and Cr 's on reproductive potential of bacteria and removal of these metals and more oxygen environment provided by vgb gene causing a more positive growth and reproduction effects were observed.
