The effect of rosmarinic acid to the complications in the experimental alzheimer model

Abstract (EN)

Alzheimer?s Disease(AD) is a common type of dementia characterized as a progressive defectiveness in cognition and behaviour. The aim of this study was to assess the effects of antioxidant and antiinflamatory properties of rosmarinic acid to the complications of AD by cognitive and biochemical parameters. Alzheimer model induced with administration of D-galactose to ovariectomized rats by using synergistic effects of estrogen deprivation and oxidative stress.In our work, 100 wistar rats are divided randomly into five groups; Control(C), Sham(Sh),rosmarinic(R),ovariectomy+D-galactose+rosmarinic(ODR), ovariectomy+D-galactose(OD) groups. OD and ODR groups were ovariectomised and administered 80 mg/kg/day D-galactose by injection, ODR and R groups were received 50mg/kg/day rosmarinic acid via gavage for 60 days. At the end of the experimental period behavioral tests were carried out and brain tissues were extracted. Then TBARS test for lipid peroxidation and ELİSA test for PGE2 was employed.Increased TBARS values was detected in Alzheimer group compared to other groups and RA improved the cognitive functions in ODR group by attenuating the lipid peroxidation. PGE2 levels were higher in OD group than in other groups and RA treatment inhibited this elevation in ODR rats. Histopathological alterations and Aß depositions were observed in the OD group. Normal cell structure and lower Aß depositions were observed in the ODR group.In conclusion, our study clearly showed that lipid peroxidation, inflamation and cognitive deficit increased in OD group and rosmarinic asid had curative effect on this parameters. And also, this new Alzheimer model can be use for other researches about AD.
