Investigation of the effects of fasudil and memantine in experimental dementia model

Objective: In this study, fasudil and memantine, which provide benefits in Alzheimer's disease by blocking the Rho / Rho Kinase pathway and Glutamate NMDA receptors when used alone, were used together against scopolamine-induced experimental dementia and their possible synergistic interactions were investigated. Materials and Methods: 56 male BALB / c mice, 3-4 weeks old, were randomly divided into 7 groups, 8 in each group. Gcontrol: 1 ml 0.9% saline ip, Gskop: 3 mg / kg Scopolamine ip, Gfas: 10 mg / kg Fasudil ip, Gmem: 3 mg / kg Memantine ip, Gskop + fas: 3 mg, once a day for 14 days Scopolamine ip / kg and Fasudil ip 10 mg / kg, Gskop + mem: 3 mg / kg scopolamine i.p. and + 3 mg / kg Memantine ip and Gskop + fas + mem: 3 mg / kg Scopolamine i.p. and + 10 mg / kg Fasudil i.p. + 3 mg / kg Memantin i.p. applied. Behavioral experiments were carried out to determine cognitive function. Levels of markers such as ABETA and BDNF were measured in the brain tissues of the subjects. Results: Levels of biochemical markers in scopolamine group were found to be different compared to other groups. This finding suggests that Fasudil and Memantine cause positive changes on synaptic plasticity. Conclusion: The ROCK signaling pathway, which causes changes in cellular responses on hippocampal learning and memory, is considered a therapeutic target in the pathogenesis of neurodegenerative diseases.
