The effects of advanced glycation end products on periodontal tissue damage in experimental diabetes and periodontitis-induced rats and evaluation of the therapeutic effects of vitamin C application

Abstract (EN)

Aim: The aim of this study was to evaluate the effects of local vitamin C treatment on tissue AGE, IL-6, 8-OHdG, MMP-8; serum levels of CTX; and alveolar bone loss in experimental periodontitis and / or diabetic rats. Material and Method: Animals were divided equally into five groups: 1) Control (C), 2) Experimental Periodontitis (P), 3) Experimental Diabetes (D), 4) Experimental Diabetes - Experimental Periodontitis (D+P), 5) Experimental Diabetes - Experimental Periodontitis - Lokal Vitamin C (D+P+LvitC). Periodontitis was induced by ligature placement in the right mandibular first molar teeth and diabetes was induced by streptozotocin injection. At the end of the experiment, serum samples were obtained and the level of CTX was analyzed. AGE, IL-6, 8-OHdG, MMP-8 were evaluated immunohistochemically and stereologically in gingival tissues. Levels of alveolar bone loss were determined using measurements performed on histological slices. Results: Serum CTX levels decreased significantly in D+P+LvitC compared to D+P group. IL-6 and MMP-8 positive cells density decreased in the D + P + LvitC group compared to the D + P group but not significant, 8-OHDG and AGE positive cell density decreased significantly were found. Serum CTX levels and IL-6, MMP-8, 8-OHdG, AGE positive cell density in the gingival tissues were significantly increased in the D + P group compared to the control group. Alveolar bone loss and attachment loss increased in the D + P group compared to groups outside the treatment group and decreased in the treatment group. Conclusion: In experimental periodontitis and diabetes rat model, benefical effects of vit C treatment on the destruction of tissue triggered by inflammation, oxidative stress and AGE accumulation observed in periodontal tissue and vitamin C may be an immunomodulator and antioxidant that can be applied locally in the treatment of periodontitis to help control glycemic showed.
