Effectiveness of low and high dose ganoderma lucidum improvements in the late period of experimental intraabdominal sepsis

Background: Intra-abdominal infections are common surgical problems and carry high morbidity and mortality risk. According to degree of host response, clinic spectrum changes from sepsis to MODS. There is no safe and realiable single therapuetic agent for management of septic shock. In this experimental study, we aimed to analyse the healing effects of low and high dose Ganoderma lucidum on septic parameters and organ injury during late sepsis. Materials and Methods: Thirty-two wistar albino rats wighing 250-300g were divided into control, septic, low dose GL treatment and high dose Gl treatment groups. The control group underwent simple laparatomy and a sham operation. Only cecal ligation and puncture was performed in septic group. Low dose GL treatment animals were given orally GL (5mg/kg daily dose), high dose GL treatment animals received orally GL(250mg/kg daily dose) one week before CLP. Twenty four hour after CLP, blood samples were collected from all groups to measure trombocyte, leukocyte hemoglobine, IL-1,IL-6 and TNF-α levels. Additionally intestinal, liver and lung tissues were examined for histopathologic changes. Results were statistically analysed by using tukey post hoc, Kaplan-Meier and Man Whitney U tests an p values <0.05 were accepted as significant. Results: Trombocyte levels were found significantly low and leukocyte, IL-1,IL-6 and TNF levels were found significantly high in septic groups compared with control groups. The healing effects of GL treatment on leukocyte and Il-1 levels were dose dependent. Septic insult resulted in significant changes in intestinal tissue (villous atrophy, tip necrosis, leukocyte infltration), liver tissue ( sinusoidal changes and leukocyte infiltration of portal area) and lung tissue( alveolary wall thickness, leukocyte infiltration). Compared with the septic group, histopathologic changes were significantly limited in GL treatment groups. Conclusions: CLP is an eligible experimental model to identify the effects of septic insult. GL has considerable (partially dose dependent) healing effects on septic parameters such as trombocytopenia and leukocytosis. GL treatment has also healing effects on organ injury due to sepsis. These healing effects of GL can be explained by its anti-oxidative, anticoagulan, anti inflammatory and immunmodulatory features.
