The healing effect of trimetazidine on ischemic tissue damage in experimental carbonmonoxide poisoning Objective: This study was planned with the hypothesis that trimetazidine (TMZ), which is known to have strong antioxidant and cytoprotective properties, could reduce the ischemic damage in vital organs such as liver, brain and heart of rats due to experimental carbon monoxide (CO) poisoning. Method: In this study, 36 mature female Spraque Dawley rats were used. Until the day of the study, the rats were kept in standard cages at 22 ºC room temperature, given water and standard rat food. In the experimental procedure, CO and air were given to the rats placed in the bell jar. Rats were divided into 6 groups. Group 1 (Sham), group 2 (TMZ + room air), group 3 (CO + 1mL SF), group 4 (CO + TMZ 5mg/kg), group 5 (CO + TMZ 10 mg/kg), group 6 ( CO + TMZ 15mg/kg). The determined protocol was applied intraperitoneally every 12 hours for 2 days. After the rats were anesthetized 48 hours after the beginning of the experimental protocol, liver, brain and heart tissues were removed and histopathological changes were examined. Results: A statistically significant improvement in myocardial cell infiltration and vascular congestion was observed at 15mg/kg TMZ dose application (p=0.035). A statistically significant healing effect was also detected in TMZ dose applications at 5 mg/kg, 10 mg/kg and 15 mg/kg levels in brain degeneration due to CO poisoning (p<0.001). When the damage occurring in the liver was evaluated, it was determined that 15 mg/kg TMZ dose application reduced sinusoidal congestion statistically significantly (p<0.001). Conclusion: In the experimental CO poisoning model created in rats, it was determined that TMZ administration at a dose of 15 mg/kg had a healing effect on brain, heart and liver damage caused by CO poisoning.
