Compare the effect of topical bevacizumab and sorafenib in experi̇mental corneal neovascularization

Neovascularization is necessary for physiological ingrowth and homeostatics but it can threat visuality if occur in transparent cornea. The purpose of this study is to compare the inhibitor effect of topical bevacizumab and sorafenib and to find effective doses of sorafenib. Six group were formed and each group had seven Albino Winstar rats. All right corneas of rats cotherized with silver nitrat except control group. Control group did not treated. In sham group three drops of saline were dribbled in a day, in group 3, topical bevacizumab 5 mg/dl was dribbled three times a day, in group 4, topical sorafenib 2.5 mg/dl was dribbled two times a day, in group 5, topical sorafenib 5 mg/dl was dribbled two times a day and in group 6, topical sorafenib 7.5 mg/dl was dribbled two times a day through 1-7 days. Corneal photographers were taken in eighth day and percentage of neovascular area in cornea were calculated. After rats were decapitated, all corneas were taken with 4 mm punch. All corneas were diveded four equal parts and in one part level of VEGF corneal immune staining was evaluated semiquantitively. In other parts VEGF level's were evaluated with ELISA. In one part one part level of immune staining PDGF was evaluated semiquantitively and in the other part level of PDGF was evaluated with ELISA. The percentage of neovascular corneal area was statistically lower in treatment groups than sham group (p<0.05). Despite the density of VEGF immune staining were lower in all treatmet groups than sham group, there were no statistically difference with control group. The level of VEGF ELISA in groups 3,5 and 6 statistically were lower than sham group (p<0.05). There were no statistically difference in any groups in level of PDGF ELISA. The level of PDGF ELISA in groups 3,5 and 6 statistically were lower than sham group (p<0.05). Topical administration of bevacizumab and sorafenib are effective in inhibit corneal neovascularization. The effect of sorafenib looks like dose-dependent. Using in low doses and applying two times a day posology are the important advantage of sorafenib.
