Abstract (EN)

Agomelatine is a novel ‎antidepressant. It enhances cognitive functions, memory, and sleep. It also holds potential ‎benefits in neurodevelopmental disorders, where it protects against oxidative stress and inflammation. However, the ‎protective capabilities of agomelatine against LPS-induced damage in BV2 ‎microglial cells remain unexplored. This study aimed to explore the ‎ potential effect of agomelatine on immunomodulatory properties of BV2 cells ‎in neurodegenerative diseases.‎ Literature ‎reported various agomelatine concentrations used in in vitro studies. Agomelatine was tested across various concentrations to acknowledge these variabilities, ‎including doses from 2 up to 200 µM on BV2 cells. Similarly, the ‎sensitivity of BV2 to LPS can also vary, leading to differing responses and toxic effects. Therefore; different ‎concentrations of LPS (ranging from 0.001 to 2 µg/ml) were also tested. The experimental design ‎involved using MTT assay to investigate agomelatine's protective properties ‎against LPS toxicity on BV2 cells.‎ Finally; the doses determined to be used were 25µM and 50µM for agomelatine and 1µg/ml for LPS. Statistical analysis showed that agomelatine has a dose-dependent effect on BV2 cell viability, and subjecting the BV2 cells to agomelatine for a prolonged duration decreased their viability. LPS at 1 μg/ml significantly ‎decreased viability compared to the control group (P < 0.001), while adding ‎agomelatine at both 25μM and 50μM significantly mitigated the toxic effects of LPS (P < 0.001). DMSO groups showed significantly lower cell viability than the control group, indicating a potential toxic effect. ‎ In conclusion, agomelatine has shown a potential protective effect against LPS-induced toxicity. Despite this potency, prolonged and DMSO negative effects should not be ignored.
