Abstract (EN)
The vast mojority of dementia patients (about 60-70%) suffer from Alzheimer disease. The Alzheimer is a progressive neurodegenerative disease with specific features like senile plaques (SP), neurofibrillary tangles (NFTs) and synaptic loss. With the second highest treatment cost of AH after cancer, there is no effective treatment. In finding a cure for this disease, I performed an experimental rat model and used as a effective on brain and especially hippocampus, oxidant selector the kolinotoksin and etilkolin mustard aziridinium ion (AF64A). In this study; we aimed to find the damage level caused by the the alcohol, inducing the cerebral atrophy and neurological anomally; and to determine the elimination and/or recession of this damage by the Piperine and Betaine as a antioxidant. The effect into mitogen active protein kinases (MAPK-1) in hippocampüs tissue, blood serum sample via mRNA level and the behaviour of the animals with using morris water maze test were searched. In this study; 60 Sprague-Dawley male rat with each 3-3,5 months old and 280-320 g weight were used. The groups were performed like this; Group 1 as the control group (n=6), Group 2 performing experimental dementia with alcohol (n=18), Group 3 performing experimental dementia with AF64A (n=18), Group 2a the control Group (only alcohol n=6), Group 2b for treatment the Piperin applying (n=6), Group 2c for treatment the Betain applying (n=6), Grup 3a the control groub (just AF64A n=6), Group 3b for treatment the Piperin applying (AF64A+Piperine n=6), Group 3c for treatment the Betain applying (n=6). From this study; we obtained expected results from as a oxidant effective of the alcohol and AF64A application on the basis of the patholojical and behavioural test in hippocampus tissue sample of blood serum, liver and hemolisat. Also, it was observed the decrease and downgrade success of Betain and Piperine into detrimental effect of these substance. Especially, the significant antioxidan effect of Piperidine was determined from statistical, behavioural, patological, blood and genetic analysis.
