Abstract (EN)
Nonalcoholic steatohepatitis is part of the spectrum of nonalcoholic fatty liver disease, a condition becoming increasingly recognized worldwide due to its prevalence in obesity, diabetes, and insulin resistance syndrome. The pathological picture bears a striking resemblance to that of alcohol-induced liver injury, but it occurs in individuals who deny a significant history of alcohol ingestion. Nevertheless, there is yet no definitive therapy that prevents it evolving through fibrosis and cirrhosis. Genistein (4?, 5, 7-trihydroxyisoflavone), a phytoestrogen found in soybeans, has a variety of pharmacological features including anticancer, antioxidant and anti-inflammatory actions. The objective of the present study was to investigate the preventive and therapeutic role of genistein in an experimental model of NASH induced by a high fat diet (HFD).A total of male Sprague-Dawley rats weighing 200-250 gr. were used in the study and rats were divided into 5 groups. Group 1 (n=10, control group; 100 ?/day 1.25% subcutan for 6 weeks); Group 2 (n=10, control group; 0.2 mg/kg/day genistein subcutan for 6 weeks); Group 3 (n=10, HFD group; 100 ?/day 1.25% subcutan for 6 weeks); Group 4 (n=10, HFD + genistein group; 0.2 mg/kg/day genistein subcutan for 6 weeks); Group 5 (n=10, NASH + genistein group; 0.2 mg/kg/day genistein subcutan for 6 weeks). After 6 weeks, rats were decapitated; blood samples obtained for biochemical parameters and liver samples for histopatholigical investigation.In this study, spesifical parameters serum TNF-alfa, IL-6, TGF-beta 1 and insulinomimetik agent Visfatin levels were measured and the effects of genistein were evaluated on these parameters.In this study; genistein was found to improve liver function tests, decrease insulin resistance, TNF-alfa, IL-6 levels, and prevent the high fat diet induced induced liver injury. In conclusion; especially genistein and consuming more food products containing isoflovanes appears to be promising in the treatment of NASH patients.
