Protective effects of carvacrol and exercise in an experimentally induced model of epilepsy with penicillin

Abstract (EN)

Epilepsy is a chronic neurological disease characterized by recurrent seizures as a result of an imbalance in neuronal stimulation. Antiepileptic drugs often cause unwanted symptoms that can affect the central nervous system. There are no studies examining the potential beneficial effects of the combined use of carvacrol and exercise on diseases affecting the nervous system. In this study, the combined effects of carvacrol and exercise on epileptic seizures were investigated with ECoG recordings. In the light of this information, it was aimed to examine whether carvacrol and exercise have a protective effect on the epilepsy model to be experimentally created with penicillin-G, with ECoG records for 8-10 weeks. Wistar albino male rat weighing 250 g was used. A total of 49 male rats were divided into seven different groups (n=7). Group-1 epilepsy model, Group-2 Carvacrol (75 mg/kg orally)+Epilepsy model, Group-3 Carvacrol (75 mg/kg orally)+ Exercise(6 days/week-6 weeks)+Epilepsy model,Group-4 Exercise(6 days/week-6weeks)+Epilepsy model,Group-5 DMSO group(6 days/6weeks orally),Group 6 Carvacrol (75 mg/kg oral)+Exercise(6 days/week-6 weeks)+diazem+Epilepsy model,Group 7 diazem+Epilepsy model and experimental epilepsy model in all groups was created with Penicillin-G (500 IU 2.5 µl, ic) injection. Basal activity, latency, spike and amplitude measurements were recorded for 2 hours with ECoG recordings and then analyzed. According to the findings, a significant effect of the combination of exercise and carvacrol on epileptiform activity latency was observed, and significant results were observed in terms of spike wave frequency and amplitude.(p <0.05). Then, hippocampal neuronal structures in the brain tissues of 48 dissected animals were stained with Hematoxylin-Eosin after tissue follow-up. Histopathological findings of CA1, CA2, CA3 regions were recorded and evaluated. Neurons in the pyramidal layer were evaluated, and histopathologically when exercise and carvacrol were applied together, it has been observed that it reduces degeneration in terms of .(p <0.05) vii KEYWORDS: Carvacrol,Epilepsy,Exercise
