Evaluation of necrostatin-1 effect on cerebral vasospazm after experimental subarachnoid hemorrhage model

Abstract (EN)
Objective: In this study, we investigated the effects of necrostatin-1(NEC-1) on subarachnoid hemorrhage (SAH) induced vasospasm in a rat model. Material and Method: 56, male Wistar albino rats were randomly divided into six groups. Group 1 (control), group 2 (sham), group 3 (decapitation one hour after SAH), group 4 (decapitation 48 hours after SAH), group 5 (NEC-1 given 15 minutes before SAH and decapitation one hour after SAH), group 6 (NEC-1 24th hours after SAH and decapitation after 48th hour). NEC-1 was applied intrasisternal with a 1 uq in dimethyl sulfoxide (DMSO) is resolved at a dose of 2.6 uq. After decapitation, basilar arteries cross sectional areas and wall thicknesses were determined histopathologically by stereological methods. Results: NEC-1 which was applied before SAH was found statistically significant to be preventive on vasospasm following SAH. Arterial wall thicknesses were found to be statistically increased only in the SAH group, but not statistically significant in the normal group, the sham group and the NEC-1 treated groups. Conclusion: The findings which are obtained from this study demonstrates that NEC-1 may be preventive in vasospasm after SAH in rats. Further studies are needed for clinical use of this agent.
