Investigation of the effects of melatonin application at experimental tonic-clonic epilesy model

The purpose of this study is to examine of central nervous system changing parameters, metabolic changing parameters and peripheric central sysytem changing parameters between control group, epileptic group whhich is induced by Pentylenetetrazole(PTZ) and melatonin treatment groups(25mg/kg and 100 mg/kg). The examinations were done both intragroups and intergroups. 40 wistar albino rat that are male and 12–14 week old divided into 4 groups(Kontrol, PTZ, MEL25, MEL100). Only solvent was injected to rats from Kontrol group and 35mg/kg dose of PTZ was injected to rats from PTZ group every other day total 23 days and 12 injections. 25mg/kg melatonin+35mg/kg dose PTZ was injected to rats from MEL25 group and 100mg/kg melatonin+35mg/kg dose PTZ was injected to rats from MEL100 group. Melatonin was injected half hour before PTZ injection and after PTZ injection rats were monitored for one hour. All doses injected intraperitoneally. The parameters that we looked for were; body weight(gram, every PTZ injection days), blood glucose levels(mg/dl, the first and last day), rectal body temperature(Co, the first,11th and 21th days ), histologically cerebellum evaluation(4X and 10X microscobic photograph, last day after decapitation), hot plate nociceptive test(second, 19th day), epileptic seizure power scale(scores, every PTZ injection days ), t maze open field escape(second, 21th day), t maze food finding(second, 21th day). We found statically significant data for rectal body temperature, blood glucose levels, seizure scores and t maze open field escape. However there is no significant data for the tests of body weight, histological evalution of cerebellum, hot plate nociceptive test and t maze food finding test(Significans value= p<0,05). Results were evaluated as both intragroup and intergroup data. Statical test that we used are; one sample t-test, paired t-test, unpaired t-test, Mann Whitney U test, Kruskal Wallis test, One Way ANOVA test. Our study showed that, melatonin has protective and healing power againist physiological changes that caused by epileptical seizures.MEL100 group data is much more significant when these data compared with the data from MEL25 group; this means melatonin has different protective and healing power at different doses. Our study is important to contains results from metabolic, neurologic and mood data together.
