Investigation of the relationship between GHRL (rs4684677), FTO (RS8044769) and PGC-1α (rs8192678) polymorphisms in diabetic patients

Abstract (EN)

Diabetes is a chronic group of metabolic disorders that generally presented with hy-perglycemia as a result of insulin synthesis defects due to multifactorial causes in beta cells in the Langerhans islets of the pancreas. In the development of diabetes, genetic predisposition is as important as environmental factors. As a result of polymorphism studies in diabetic patients, many genes have been found to be associated with the deve-lopment of diabetes. In our study, we aimed to represent the relationship between diabe-tes and certain variants of the GHRL, FTO and PGC-1α genes, which are generally as-sociated with diabetes and obesity, by comparing the patient and control groups. 100 Type 2 DM patients and 94 healthy volunteers were enrolled in our study. GHRL (rs4684677), FTO (rs8044769) and PGC-1α (rs8192678) gene polymorphism studies were performed by Real-Time PCR method after DNA isolation. One Way ANOVA test, Post-Hoc and Kruskal-Wallis tests were applied using SPSS in order to show the statistical differences among two groups. The carriers of TT allele for the FTO (rs8044769) variant and the GG allele for the PGC-1α (rs8192678) variant were found more frequently in the patient group, while the GHRL (rs4684677) did not differ between the patient and control groups. For PGC-1α (rs8192678) variant in the patient group, glucose and BMI levels were observed to be significantly higher in carriers of the GA genotype than those with the GG genotype. There was no statistical difference in the distribution of GHRL (rs4684677) alleles according to biochemical profiles. Further studies are needed to explain the relationship of related genes with diabetes.
