Toxic effect of dimethoate on rate testicular tissue and protective role of ferulic acid

Abstract (EN)
Chemical pesticides, pesticides, disease, pests and weeds are the most preferred products of the manufacturers because of their high impact in a short time. Uncontrolled and unconscious use of pesticides may cause negative effects on all living organisms, especially human health, due to their residues in food. Dimethoate used in this study is an organophosphate pesticide. Ferulic acid is known to have antioxidant properties. In this study, subacute dimethoate was given to rats in two different doses as low (1/100 LD50) and high dose (1/10 LD50) by oral gavage. Ferulic acid, an antioxidant, was also administered by oral gavage at a dose of 30 mg / kg each day to examine the effects of testicular tissues on rats. For this purpose, the levels of glutathione peroxidase (GPx), superoxide dismutase (SOD), glutathione-S-transferase (GST) catalase (CAT), and lipid peroxidation marker malondialdehyde (MDA) were investigated to measure oxidative stress effects. Histopathological examinations were performed with a light microscope. For this purpose, experimental animals were divided into 6 groups. The control groups are ferulic acid, low dose dimethoate, high dose dimethoate, ferulic acid and low dose dimethoate, ferulic acid and high dose dimethoate. Ethics committee approval was received from Gazi University Animal Experiments Local Ethics Committee for this experimental study (G.U. ET-17.004). Following administration to animals, testicular tissues were taken from rats in each group, frozen in liquid nitrogen and stored in a -80°C freezer. Antioxidant enzyme and MDA levels were measured spectrophotometrically according to various methods. According to the results, no statistically significant difference was observed in all parameters in testicular tissues of rats who were administered control and ferulic acid. When the control and ferulic acid and low / high dose dimethoate were compared, it was observed that there was a statistical significant increase in SOD, CAT, GPx, GST enzymes and MDA levels. There was a statistically significant increase in ferulic acid and low and high dose dimethoate groups when compared to the control group, while a statistically significant decrease was observed in low and high dose dimethoate groups. As a result of histopathological examinations, while some toxic changes were observed in dimethoate applied groups, less toxicity was observed in ferulic acid applied groups. As a result, it is possible to say that ferulic acid has a partially healing role on the oxidative damage caused by dimethoate.
