Investigation of the effects of asprosin administration on energy metabolism-related hormones in female rats

Abstract (EN)
For the sustainability of metabolism, which is one of the basic components of life, the body must have an internal balance. This balance consists of a series of biological processes involving energy intake (nutrient intake) and energy consumption. The coordinated functioning of peripheral and central neuronal inputs, including hormones, peptides, and neurotransmitters, is required to regulate energy homeostasis and maintain a balance between food intake and energy expenditure. Impaired energy metabolism leads to many diseases such as obesity, although it reduces the quality of life. Adipose tissue, which is one of the main energy sources, is an effective organ in the regulation of metabolism. The function of asprosin administration, which is an adipokine secreted mainly from adipose tissue, in energy metabolism is still unknown. In this direction, our study was carried out to determine the effects of asprosin administered to female rats on some hormones related to food consumption and energy metabolism. In our study, 24 female Sprague-Dawley rats, 21 days old and weighing 35 ± 2 grams, obtained from Fırat University Experimental Research Center (FUDAM) were randomly selected and divided into two groups as control and asprosin groups (n=12). From the 21st day onwards, female rats regularly administered asprosin between 13:00 and 15:00 for an average of 8 weeks; It was aimed to reveal the possible effects of asprosin administration on feed-water consumption, weight change and hormones effective in energy metabolism in rats by daily monitoring of eating-drinking behaviors and weight changes. Asprosin (500ng/kg) daily was administered intraperitoneally to the animals in the asprosin group for an average of 90 days from the 21st day after birth. Similarly, physiological saline was administered to the control group (1ml/kg). Feed, water consumption and weight changes were monitored between 13:00 and 15:00 for an average of 8 weeks. Experimental groups were decapitated at the end of the 90th day without being anaesthetized. Asprosin, ghrelin, leptin, glucagon, glucose, insulin, follicle stimulating hormone (FSH), thyroid stimulating hormone (TSH), growth hormone (GH), kispeptin corticosterone, inhibin A, inhibin B levels were investigated in blood samples taken after decapitation with ELISA kit. . It was observed that there was no statistically significant difference in the feed, water consumption and weight gains of the control group and asprosin group female rats. When the serum samples were examined, a statistically significant difference was observed in serum asprosin, ghrelin, corticosterone, GH, FSH, TSH, inhibin B levels between the two groups. In biochemical examinations, no significant difference was observed in lipid profile, lipid profile (LDL), high-density lipoprotein (HDL) and Triglyceride (TG) levels
