Cervix collagens and vascular structure in diabetes and normal, young and old pregnants

Abstract (EN)

Diabetes mellitus is a heterogeneous metabolism disorder that became specific with hyperglycemia. Diabetes has negative effects on pregnancy and pregnancy has also on diabetes. Aging is a complex process where functions from cells to organs gradually decreases in all structures. Changes occurring with aging affect development and outcome of diabetes. In our study, immunohistochemical assessments were performed using collagen Type III, IV, VI and alpha actin primary markers in order to determine the possible effects of aging and diabetes in addition to pregnancy in cervix tissue known that it demonstrates vascular changes and reconstruction in collagen fibers during pregnancy. In our study, 48 female Sprague Dawley rats as 2 months old for young groups and as 8 months old for old groups were used. Rats were divided into total 8 groups as 6 subjects in each group. Single dose of 50 mg/kg STZ was applied intraperitoneally to subjects in groups with diabetes. After follow-up for 2 months following administration, pregnancy formation was achieved in subjects that will be made pregnant in diabetes and control groups. At the 14th day of pregnancy, cervix tissues of all groups were removed. Immunohistochemical and statistical outcomes were assessed. In conclusion, in our study, increase of cervix collagens in all markers in the advancing middle stages of pregnancy suggested us that they are necessary to provide continuity of pregnancy. It was of opinion that these markers may reduce when close to delivery. Increase in collagen expression with advanced age was accepted as a markers of regression of tissue with senility. It was of opinion that increase in all collagen expression in diabetic groups is in order to reduce the possibility of abortion. While alpha actin expression was accepted as an indicator of apoptosis, reduction in stroma in pregnant and diabetic groups, increase of it in smooth muscles on vascular wall suggested us the result that it carried out concurrent to increasing vascularization.
