Investigation of the effect of topical vascular endothelial growth factor (VEGF) administration on oxidative status of wound tissue in diabetic rats

Abstract (EN)
Growth factors? not to be produced enough and catabolism?s increase are main factors of the late wound healing in diabetics. Vascular endothelial growth factor (VEGF) plays role in the wound healing by inducing vessel development. In this study we aim to investigate the topical VEGF administration on oxidative status of wound tissue in diabetic rats. In our study 36 female Wistar albino rats (weight: 200-250g) were used. Rats get diabetic with streptozotocin. Dorsolateral excisional wounds were made on rats and they were randomly divided into 3 groups: Untreated group (n=12), Chitosan treated group (n=12), Chitosan+ VEGF treated group (n=12). In VEGF groups, wounds were treated topically with single daily dose VEGF-A (7ng/ml). In chitosan treated groups, wounds treated topically with equal amount blank chitosan gel. After these administrations, on the 3th and 7th days of wound healing, animals were sacrificed. NOx, TBARS, AA, GSH levels and SOD activity were measured spectrophotometrically in wound tissues. Results were expressed as mean ± standard deviation. Mean differences were compared by Anova variance analysis (P<0, 05). Topical VEGF especially decreased NOx level in early level. It increased TBARS levels in both days. Also it increases GSH and AA levels contributed to increase of wound tissue antioxidant capacity. In the light of this information VEGF administration may have altering effects on oxidative events depending on days in wound healing process.
