Investigation of the resveratrol and grape molasses effects on the hepatic enzymes and oxidative stress paramters in the rat liver injury induced by DMBA

Abstract (EN)

Resveratrol is a potent antioxidant and some fruits such as black grapes, peanut and ananas have great amount of resveratrol. Our experiments aim to investigate the antioxidant effects of grape mollesses on DMBA induced hepatic injury in terms of hepatic enzymes and oxidative stress parameters. We also want to compare the antioxidant effects of grape mollesses and resveratrol in hepatic injury. 42 female (18 weeks) Wistar Rats were used in this experiment. Rats were distrubuted to 6 groups as follow. 1. Sussame oil plus DMSO (2:3 portionally) were enjected 10 mg/kg subcutaneously (sc per day), in the control group; 2. 10 mg/kg DMBA enjected subcutane only on first and 7th days, in the DMBA group; 3. 20 % percent molassas feeding and DMBA injection, in the DMBA+Molasses Group; 4. DMBA injection subcutane only on the first and 7th days; 10 mg/kg resveratrol subcutane per day, in the DMBA+ Resveratrol group; 5. only 20 % percent molassas feeding, in the Molasses Gruop; 6. 10 mg/kg resveratrol was enjected subcutane per day, in the Resveratrol group. Rats were sacrified after the taking liver tissue and blood sample on the 10th day . ALT, AST and GGT levels were measured in serum samples and liver tissues used for measurement of total antioxidant capacity (TAS), total oxidant capacity (TOS) and 5'Nucleotidase levels. The comparision of ALT, AST and GGT levels, a significant decrease were detected in the DMBA+Resveratrol and DMBA+Molasses groups comparing to DMBA group. Beside, this decrease is more prominent in DMBA+Molasses group according to DMBA+Resveratrol group. However we found 5'Nucleotidase enzyme activity was suppressed this increasing in DMBA group DMBA+ Molasses and DMBA+Resveratrol groups. According to these results, we can claim that both resveratrol and grape mollasses have benificial effects on hepatic injury. In this study we observed that liver TAS levels was decreased in DMBA group compared with the control group. But DMBA+Resveratrol and DMBA+ Molasses groups have a significant increase in liver TAS compared with only DMBA injected group. We also observed a decrease in liver TOS levels in DMBA+Resveratrol and DMBA+ Molasses groups compared with DMBA injected group. As a conclusion, we concluded that Molasses and Resveratrol can supress for oxidative stress levels and activate the antioxidant system in DMBA induced liver injury.
