Molecular characterization of raphidascaris acus (bloch, 1779) (Raphidascarididae) based on DNA sequences

Abstract (EN)

In this study, the molecular identification of the Raphidascaris acus species (Nematoda, Ascarididae), which is protected as a material in the ethyl alcohol environment, was carried out on the 5.8S gene sequence. The 5.8S rRNA gene region sequence data confirmed the taxonomic position of the R. acus species, which was identified according to their anatomical and morphological structures. In this process, specifically designed AcusF and AcusR primers were used. 750 bp partial certain nucleotides of the gDNA (5.8S rRNA) gene was sequenced directly from each sample. No nucleotide variations were detected in the 5.8S gene sequences of the 12 R.acus samples examined in the study. In terms of the 5.8S aligned nucleotide sequences, the minimum nucleotide variations were detected between the present Raphidascaris acus isolates (MK271779-MK271790) and the two R.acus isolates (KT633862, KM047505).On the other hand, AY603537 R.acus isolate showed 4.11% difference. Our R.acus isolates differed by three Raphidascaris species (JN102362.1 Raphidascaris longispicula, FJ009682.1 Raphidascaris trichiuri, JF809816.1 Raphidascaris lophii, KR232377.1 Raphidascaris macrouri) 13.03-14.97%, GQ215514.1 Eustrongylides sp. external group 26%. In addition, the present data showed that R.acus gDNAs fixed in ethanol and preserved in ethanol for a long time were not damaged and routinely reproduced.
